Summary
Semax is a synthetic heptapeptide analogue of the ACTH(4-7) fragment developed in Russia. It has been studied for cognitive enhancement, neuroprotection after stroke, and ADHD-like conditions. Increases BDNF, NGF, and serotonin in key brain regions in animal models.
Semax is based on a fragment of ACTH — the stress hormone — but the modifications remove the hormonal effects and leave only the brain-boosting ones. It significantly increases BDNF (the brain's 'fertilizer' for neurons) and activates dopamine and serotonin systems. Used in Russian hospitals for stroke recovery. The research community uses it for focus, memory, and mood. Often described as feeling like cleaner, calmer concentration.
Mechanism of Action↓
Derived from ACTH 4-7 but modified for metabolic stability. Increases expression of BDNF, GDNF, and NGF. Activates dopaminergic and serotonergic systems. Demonstrates neuroprotective effects in ischemia models via anti-apoptotic and antioxidant mechanisms.
Half-life: ~5–7 minutes (intranasal); longer than Selank due to Pro-Gly-Pro C-terminus
Research Dosing Ranges↓
100–900 µg per dose
1–3× daily intranasal or subcutaneous
Most literature uses intranasal administration at 50–300 µg per nostril.
Reconstitution Guidance↓
BAC water: 1 mL
Resulting concentration: mg/mL depends on vial size
Dilute further for intranasal use. Standard SC reconstitution otherwise.
Storage & Stability↓
Refrigerate. Reconstituted: use within 14 days.
Known Side Effects / Contraindications↓
Mild anxiety, irritability at high doses. Generally well tolerated.

Concentration: 5 mg ÷ 3 mL = 1.67 mg/mL (1667 µg/mL)
Dose needed: 250 µg = 0.2500 mg
Volume per dose: 0.2500 mg ÷ 1.67 mg/mL = 0.1500 mL
On U-100 syringe: 150.0 µL = 15.0 units
Total doses in vial: 5 mg ÷ 0.2500 mg = 20.0 doses
