Summary

Semaglutide is a long-acting GLP-1 receptor agonist with high albumin binding that extends its half-life to ~1 week. It is one of the most extensively studied peptides in this class, with robust data across multiple outcomes including glycemic control, cardiovascular risk reduction, and body weight.

The same active ingredient as Ozempic and Wegovy. It mimics a hormone your gut releases after eating that tells your brain to stop eating, slows down digestion, and helps your pancreas control blood sugar. Once-weekly injection. The original blockbuster weight-loss drug in this class.

Mechanism of Action

Highly selective GLP-1R agonist. Stimulates glucose-dependent insulin secretion, inhibits glucagon, delays gastric emptying, and reduces food intake via central (hypothalamic) appetite signaling. The C18 fatty diacid chain enables albumin binding, dramatically extending half-life.

Half-life: ~7 days

Research Dosing Ranges
⚠ Research reference only — not medical advice, not for human use.

0.25 mg – 2.4 mg

Once weekly subcutaneous injection

Diabetic research: titrate to 0.5–1 mg. Obesity research: titrate to 2.4 mg over 16–20 weeks.

Reconstitution Guidance

BAC water: 1 mL

Resulting concentration: mg/mL depends on vial size

Add 1 mL BAC water per mg of peptide for 1 mg/mL concentration. Adjust BAC volume based on vial size listed.

Storage & Stability

2–8°C. Reconstituted: use within 28 days refrigerated.

Known Side Effects / Contraindications
⚠ Research reference only — not medical advice, not for human use.

Nausea, vomiting, diarrhea, constipation (very common), injection site reactions, increased heart rate. Potential thyroid C-cell hyperplasia signal (animal data — relevance to humans uncertain).

Concentration: 5 mg ÷ 3 mL = 1.67 mg/mL (1667 µg/mL)

Dose needed: 500 µg = 0.5000 mg

Volume per dose: 0.5000 mg ÷ 1.67 mg/mL = 0.3000 mL

Total doses in vial: 5 mg ÷ 0.5000 mg = 10.0 doses
