Summary

MOTS-c is a 16-amino acid peptide encoded within the mitochondrial genome (12S rRNA). It acts as a retrograde signal from mitochondria to the nucleus, regulating metabolic homeostasis. Research in rodents shows exercise-mimicking metabolic effects, improved insulin sensitivity, and increased longevity.

MOTS-c is unusual — it's one of the only known signaling molecules encoded in mitochondrial DNA (your cellular power plants) rather than the nuclear genome. When cells are stressed metabolically, MOTS-c is released as a signal to the rest of the cell and body to ramp up glucose uptake and fat burning — mimicking what exercise does. Old mice given MOTS-c showed significantly improved metabolic health.

Mechanism of Action

Activates AMPK (AMP-activated protein kinase) and inhibits the folate cycle and de novo purine synthesis. Translocates to the nucleus during metabolic stress where it regulates gene expression. Increases glucose uptake in muscle, improves mitochondrial function.
