Summary
Melanotan II is a synthetic cyclic heptapeptide analogue of α-MSH that activates multiple melanocortin receptors (MC1R, MC3R, MC4R, MC5R). In research it has been studied for photoprotective tanning, sexual dysfunction (libido/erections via MC4R), and appetite suppression.
Like MT1 for tanning, but it also crosses into the brain and activates receptors that drive sexual arousal and suppress appetite. This is the peptide PT-141 was derived from. The multi-receptor activity makes it more potent but also harder to manage — nausea and spontaneous erections are common early side effects. Requires UV exposure for the tanning to activate after loading.
Mechanism of Action↓
Non-selective melanocortin receptor agonist. MC1R: melanin synthesis. MC3R/MC4R: CNS effects including sexual arousal, appetite suppression, autonomic changes. Crosses the blood-brain barrier, producing stronger central effects than MT1.
Half-life: ~1–2 hours
Research Dosing Ranges↓
0.25–1 mg per injection
Loading: daily for 1–2 weeks with UV exposure. Maintenance: every 2–3 days.
Start at 0.25 mg to assess response; nausea and spontaneous erections are common at higher doses. UV exposure (tanning bed or sun) activates melanogenesis after loading. Once desired pigmentation is reached, reduce to maintenance dosing.
Reconstitution Guidance↓
BAC water: 3 mL
Resulting concentration: mg/mL depends on vial size
Add 3 mL BAC water. Keep doses minimal to manage side effects.
Storage & Stability↓
Refrigerate. Protect from light. Reconstituted: use within 28 days.
Known Side Effects / Contraindications↓
Nausea, facial flushing, spontaneous erections, yawning, darkening of moles/nevi, increased libido. Mole monitoring recommended. Potential cardiovascular effects at high doses.

Concentration: 10 mg ÷ 3 mL = 3.33 mg/mL (3333 µg/mL)
Dose needed: 500 µg = 0.5000 mg
Volume per dose: 0.5000 mg ÷ 3.33 mg/mL = 0.1500 mL
On U-100 syringe: 150.0 µL = 15.0 units
Total doses in vial: 10 mg ÷ 0.5000 mg = 20.0 doses
