Summary
IGF-1 LR3 is a synthetic variant of Insulin-like Growth Factor-1 (IGF-1) with an Arg3 substitution and 13-amino acid N-terminal extension that reduce binding to IGF binding proteins (IGFBPs), dramatically extending its half-life and bioavailability compared to native IGF-1.
IGF-1 is the hormone that carries out most of growth hormone's muscle-building effects in tissues. Native IGF-1 clears from the blood in about 10 minutes — this modified version lasts 20–30 hours because it doesn't get grabbed and inactivated by the proteins that normally neutralize it. It directly stimulates muscle cell growth and protein synthesis. Serious hypoglycemia risk — always have carbohydrates available.
Mechanism of Action↓
IGF-1 receptor (IGF-1R) agonist. Activates PI3K/Akt and MAPK pathways to promote protein synthesis, cell proliferation, and anti-apoptosis. Reduced IGFBP affinity allows sustained tissue exposure. Anabolic in skeletal muscle; promotes satellite cell activation and hyperplasia.
Half-life: ~20–30 hours (vs ~10 minutes for native IGF-1)

20–100 µg per day
Once daily intramuscular or subcutaneous injection (bilateral injection sites used in some protocols)
Higher doses associated with hypoglycemia risk. Start at 20–40 µg. Must have carbohydrates available post-injection.
Reconstitution Guidance↓
BAC water: 1 mL
Resulting concentration: 100 µg/mL (for 100 µg vial) or 1 mg/mL (for 1 mg vial)
Reconstitute in sterile acetic acid (0.1 N) then dilute in BAC water or saline. Some vials come pre-acidified.
Storage & Stability↓
Freeze at -20°C before reconstitution. Reconstituted: refrigerate, use within 3 days.
Known Side Effects / Contraindications↓
Hypoglycemia (serious), jaw/extremity growth with chronic high use, carpal tunnel, potential increase in cancer risk with chronic use (IGF-1R signaling). Requires careful dosing.
